We have MAILsweeper 5.2c running on a Domino 6 server with 2 mail boxes and it works fine. Much more powerful than the R6 filters though we use them too but for different purposes.
Basically R6 filters are too difficult to fine tune to do things like spotting pr0n and the like reliably.
For example, at one stage I had an R6 filter that sidelined any message with FW: FW: or Fwd: Fw: (and several permutations along those lines) in the subject line. This was intended to stop people who, on receiving those irritating "guardian angel" mass mailings, read all the way to the bottom and, when confronted with the exhortation "now send this to all your friends and something really good will happen to you in 4 minutes", actually did so. It worked too, for a while and then I started grabbing legitimate mail too - the collateral kill rate was just too high.
By contrast, MAILsweeper handles this sort of stuff much more sensibly, can handle attachments (e.g. block MP3, MPEG and so on based on signatures not filenames) and integrates with most virus scanners including my favourite (Trend ScanMail).
Just one problem. Registered version doesn't recognise R6 as a supported platform, so when I enter our license key and start the server, all the MAILsweeper processes start up and die immediately with an error to the effect of "Invalid Domino Version" or some such. I am having to run in 30 day evaluation mode pending a fix to this, but it does still work.
